Protein aggregation - Implications in biomedicine and human health

Protein aggregation is one of the grand problems of biophysics. It is highly complex, sensitive to initial conditions, operates on a wide range of timescales and its products range from dimeric proteins to macroscopic fibrils. Unwanted aggregation, for instance, has implications in biotechnology, where protein aggregation leads to reduced yield in bioprocessing, and human health, where aggregation-based diseases such as Alzheimer’s, Parkinson’s, Huntington’s and Type II diabetes affect millions of people. Hence, an understanding of the molecular mechanisms underlying protein aggregation is of great relevance in diverse research fields such as medicine, pharmacy, food science, bionanotechnology and industrial biotechnology.
The challenge is when proteins are exposed to  various types of stress, including high concentrations, variable temperatures, pH extremes and varying ionic strengths that can lead to aggregation. To overcome these challenges, one approach is to formulate the proteins with stabilizing molecules, so-called excipients, that prevent irreversible protein aggregation. Beyond the general trends, the role of excipients based on small organic molecules in dictating protein stability is unpredictable. In order to understand protein aggregation on a molecular level, the project involves computational and experimental approaches.
Isothermal Chemical Denaturation (ICD) will be used to simultaneous measure stability and aggregation of proteins under various physico-chemical conditions. ICD is unique in being reversible for most protein systems, hereby enabling determination of thermodynamic stability, prediction of protein aggregation and estimation of dissociation constants for excipients. Complementary, molecular dynamics simulations will be performed to study (on the molecular level) the effect of selected physico-chemical conditions and excipients on protein stability and aggregation.
