Taxonomic characterization of novel lactic acid producing bacteria

In 2018, Novozymes together with Astra ran the “Masseeksperimentet 2018”, where 25,000 Danish school students collected samples of plants, tested for presence of lactic acid producing bacteria (LAB). From 11142 samples submitted to Novozymes 3906 unique LAB isolates were identified. Most of the strains belong to already known species of LAB, but subset of these were identified by whole genome sequencing. Sequence data indicate that these represent at least 10 novel species of LAB.

In collaboration with DTU food Novozymes would like to characterize LAB strains of the novel specie to make a formal description of these species. Formal description of novel bacteria requires a publication in “International Journal of Systematic and Evolutionary Microbiology”. The requirements for such publication is relatively well described. The description of novel bacterial species is based on physiological and biochemical characterization of the strains as well as data indication the genomic distance to the closest related valid described bacterial species. The formal genetic basis for species distinction was based on 70% DNA/DNA homology, which when comparing whole genome sequencing translate to 95% pairwise ANI.

In this project, the novel isolates are divided into taxa based on pairwise ANI analysis. This may be supplemented with additional bioinformatic characterizations. The novel isolates are preserved in a collection in a format suitable for conducting the various test in a semi-automated manner. The assays for this need to be set up and validates using relevant benchmark strains. (Type strains of the closest relatives).
