Are you looking for an exciting, multidisciplinary project for your master’s thesis? Do you have a background in biotechnology, molecular biology, microbiology, biochemistry, metabolic engineering or synthetic biology? The Systems Environmental Microbiology group at the Novo Nordisk Foundation Center for Biosustainabiliy, DTU (www.sem-cfb.com) is looking for a highly motivated, passionate student for a master’s thesis project starting as soon as possible.

 

The project revolves around the degradation and valorization of xenobiotic substances and the production of new-to-Nature chemical compounds. You will work with the soil bacterium Pseudomonas putida - an ideal biotechnological platform due to its fast growth, low nutrient demands and an extremely flexible metabolism that enables it to adapt to many different physicochemical conditions and to high concentrations of toxic substances. You will work with modern molecular tools to engineer P. putida into an efficient production host. You will furthermore learn how to design synthetic biology-guided metabolic engineering strategies to select for desired traits in a biotechnological chassis.

 

Based on the progress within the project, the employed methods and key topics comprise:

·       state-of-the-art molecular biological techniques for the engineering of bacterial cell factories

·       carbon metabolism of soil bacteria

·       metabolite analyses via HPLC, NMR and spectrophotometric assays

·       cultivation and characterization of bacterial strains in microtiter plates, shake flasks and bioreactors

·       enzyme engineering: rational design and directed evolution

·       use of biosensors for the in vivo detection of desired products, combined with flow cytometry and FACS
