Lactococcus cloning by Gibson assembley

Gibson assembly is probably the fastest and most versatile method for the construction of recombinant DNA molecules.
The method is simple and efficient for cloning in highly competent E. coli cells.
It is not desirable to use E. coli as an intermediary if the cloned genes affect E. coli negatively.
In this project you will develop a protocol for the direct transformation of Gibson assembly reactions into Lactococcus lactis. You will op-scale the Gibson reaction to obtain larger amounts of recombinant DNA molecules in order to compensate for the relatively lower transformation efficiency in Lactococcus lactis.
