C-reactive protein - An acute phase protein explored as a biomarker

Background: C-reactive protein (CRP) is a prominent acute phase protein in a number of species including humans. CRP is associated with the acute phase response to tissue damage as an evolutionary ancient innate systemic host defense reaction. The biosynthesis of CRP is triggered by tissue damage resulting in dramatic (>100 fold) increases in serum concentrations following shortly after injury. CRP has five identical subunits each being capable of independently binding one molecule of phosphocholine (see figure). It has innate immune modulating activities which are dependent on the binding of phosphocholine (PC) and on the structural integrity of the pentamer. Project aim: The aim of this project is to investigate modulation of CRP bioactivity by phosphocholine and synthetic phosphocholine derivatives mimicking the biologically relevant phosphocholine structures . Three types of phosphocholine derivatives, simple phosphocholine and phosphocholine with two different spacers will be studied initially (see figure). Subsequently, the binding of multivalent phosphocholine ligands will be investigated using computational methods. The binding affinities calculated using the computational methods will be correlated to binding affinity bioassays (e.g. ELISA) and furthermore correlated with bioassays measuring the subsequent pro- or anti-inflammatory activation of CRP.
