MicroScribe digitizer-probe is a device for collecting 3D coordinates by tracing an object with a tip of a mechanical pointer arm. MicroScribe is widely used for collecting 3D landmarks in geometric morphometric, a study of shape variation between organisms, usually done by collecting coordinates of landmarks and using multivariate statistics to describe the patterns within and among groups. In this project we want to challenge the MicroScribes position as the gold standard, and investigate how other data capturing techniques compare with MicroScribe when quantifying the shape and its deformations.

A number of example objects to be used in this project will be provide by Natural History Museum of Denmark. As a reference data, we will use object landmarks captured using MicorScribe. The objects will be digitized using the structured light scanner built and developed at DTU Compute, and previously used for scanning human and animal crania. For some of the objects we will collect further data using a micro CT scanner at DTU Physics. The landmark positions will be marked on digitized models by different operators. Finally, we will perform a statistical analysis to estimate the intra-observer and inter-observer reliability of landmark measurements across the different digitizing techniques.
