Lactococcus lactis cell wall proteinase

Lactic acid bacteria (LAB) are auxotrophic for several amino acids and usually depend on an active proteolytic system in order to grow in natural environments as milk. A key enzyme for many LABs is a large extracellular protease covalently linked to the cell wall. It has recently been shown that growth in camel milk pose a challenge for LABs commonly used in bovine milk, and that this challenge is linked to the proteolysis.

We have isolated Lactococcus lactis strains from camel milk and found several new variants of the cell wall proteinase prtP. The isolates show a different proteolytic pattern on caseins from camel and bovine milk. Several strains have been sequenced and the prtP gene identified. Three new versions of prtP have been identified and one of these is not covalently linked to the peptidoglycan of the cell wall.

Several projects are possible within the analysis of prtP from Lactococcus lactis. A combination of molecular genetics and biochemistry will be used to gain a deeper understanding of the function of the PrtP enzyme.

Different angles can be applied to analyze the gene and the enzyme:

  • Analysis of the function of the individual amino acid differences between the known variants of prtP by shuffling

  • Analysis of the function of individual domains of the enzyme

  • Proteomic analysis of cleavage specificity

  • Selection of mutants with altered specificity

  • Bioinformatic analysis to compare structure and specificity among G-positive cell wall proteinases
